Abstract: Endomembranedamage constitutes a significant stressor detrimental to eukaryotic cells. Tocounter this threat, cells have evolved mechanisms to repair such damage andrestore cellular homeostasis. Yet, the processes through which cells stabilize damagedendomembranes and how damaged organelles interact with and affect otherorganelle functions are not well characterized. In this seminar, I will delveinto the effects of lysosomal damage on human macrophage metabolism andelucidate the role of stress granules. These membraneless ribonucleoproteinassemblies are pivotal in providing membrane stabilization and supporting hostdefense against Mycobacterium tuberculosis, a human pathogen that exploitsendomembrane damage to sustain its survival within the host. Finally, I willdiscuss future perspectives aimed at exploiting single-organelle resolutionapproaches to understand lysosome and organelle heterogeneity and its impact oncellular function. Through a closer examination of the emerging understandingof condensates and lysosomal functions in both normal and aberrant cellularstates, we aim to uncover novel insights into human diseases and identifypotential therapeutic strategies.
